Marco Rubio: GOP Has Immigration 'Challenge' ? Peace and Freedom

Florida Sen.?Marco Rubio?told me this morning that he agreed with Los Angeles Mayor Antonio Villaraigosa, who said Tuesday that?you can?t just trot out a brown face or a Spanish surname and expect people are going to vote for your party or your candidate.?

Villaraigosa was referring to the number of prominent Hispanic Republicans slated to speak at the?Republican National Conventionthis week, but Rubio said the mayor?s message applies to both parties.

?I think what he?s saying, quite frankly, is true for both parties,? said Rubio,?who spoke to me this morning on ?Good Morning America.? ?Policies matter and, look, the Republican Party does have a challenge. We can?t just be the anti-illegal?immigration?party, we have to be the pro-legal immigration party.

?We have a broken system of immigration that needs to be fixed and we have a legal immigration problem because we have millions of people who are in this country without documents and we have to deal with that in a way that honors our legacy both as a nation of compassion and a nation of immigrants but also as a nation of laws. That?s a real challenge. That?s not an easy thing to do. We?re going to confront that not by pandering, not by making unrealistic promises but by trying to find a balanced approach to how to deal with it.?

Recent polls show that?Mitt Romney, the GOP nominee for president,?is struggling with Latino voters. Hispanics are a growing minority in the United States and a key demographic in Rubio?s home state of Florida, a crucial swing state in the presidential election. Romney has expressed views on immigration seen as being to the right of former?President George W. Bush?- who was for comprehensive?immigration reform?- while President Obama, who has much higher support among Latino voters, issued an executive order earlier this year that would?allow some undocumented immigrants legal status.

Rubio told me this morning that Romney would replace this policy.

?I think he?s going to replace it. That?s what I think you can expect from the Romney presidency and I think the obligation of some of us in the Congress is to find that permanent solution,? Rubio, 41, said. ?It was something I was working on, that I continue to work on, an alternative to the DREAM Act that allows us to continue to deal with that issue but not in a way that?s amnesty and not in a way that encourages illegal immigration in the future.?

Rubio, who is Cuban-American, is among several?Hispanic Republicans?who?have prominent roles?at the Republican National Convention this week. Rubio will introduce Romney Thursday night on the final night of the convention in Tampa, Fla., before the former Massachusetts governor formally accepts the GOP nomination for president.

?I hope to present to the people of the United States in the few minutes that I have, two things. A very unique human being, Mitt Romney, who has lived his life in a way that?s not just admirable but really a role model, irrespective of how people may feel about his policies, they may disagree with him on policies, but you look at the way he?s lived his life as a husband, as a father, as a member of his community, really a role model for younger Americans and what we should all aspire for our kids to be,? Rubio said.

?And then, also, the choice that America has between two very different views of government?s role in our economy. That?s really what this election is about. It?s not just a choice between a Democrat and a Republican, it?s a choice between two very different futures. I hope I can do that for him.?

High wind, rains lash Florida Keys as Isaac strengthens

NBC's meteorologist Bill Kearns has Isaac's latest trek.

Updated at 5:41 p.m. ET: Isaac lashed south Florida with strong winds and heavy rain on Sunday after battering the Caribbean, and officials said the tropical storm could?strengthen?into a hurricane by midweek as it heads toward the Gulf Coast.

Emergency management and law enforcement officials in Key West ordered visitors and residents to remain indoors as the storm moved through the island chain. ?You?ve chosen to remain in the Keys during this storm and the only safe place for you to be is indoors,? said Monroe County Emergency Management Director Irene Toner. ?Stay off roads and don?t go outside.?

Isaac caused weekend havoc in Cuba, where it downed trees and power lines. Before that, Isaac was blamed for seven deaths in Haiti.


Isaac was still a tropical storm Sunday evening with 60 mph winds as it moved past the Florida Keys and headed into the warm waters of the Gulf of Mexico.?Forecasters warned that Isaac could be upgraded to a Category 2 hurricane ? capable of sustained winds of 96-100 mph ? as it hits the northern Gulf Coast somewhere between Florida and Louisiana later this week.

In Louisiana, Gov. Bobby Jindal declared a state of emergency and?urged residents in low-lying areas of several southeastern parishes to voluntarily leave ahead of the storm. He said mandatory evacuations could be ordered on Monday.?The governor also activated 4,000 National Guard troops and informed?other states that Louisiana might need assistance if hit by Isaac.

New Orleans Mayor Mitch Landrieu declares a state of emergency for the city of New Orleans in advance of Tropical Storm Isaac.

"We?re all going to err on the side of being overprepared," Jindal said. He added that he may skip his speaking engagement at the Republican National Convention in Tampa, Fla., this week if his state is still threatened by the storm.

In the city of New Orleans, which was ravaged by Hurricane Katrina seven years ago,?Mayor Mitch Landrieu also declared a state of emergency.?"I'll remind everybody that we thought Katrina would be a wind and rain event," Landrieu said.

The National Hurricane Center on Sunday evening discontinued a hurricane warning for the Florida Keys and west coast of Florida, but issued a hurricane warning hurricane for the northern Gulf of Mexico coast from the New Orleans area to the Florida Panhandle.

In South Florida, three people were killed in two separate crashes due to wet roads,?reported NBC News affiliate NBCMiami.com. The first crash involved a head-on collision, which killed both drivers, and in the second, the car plunged into a canal and the driver drowned, officials said.

Dr. Rick Knabb, director of the National Hurricane Center, said forecast models ?can drive us nuts sometimes? but they play an important role for guidance.

The Florida Keys know what to expect from a big storm, and they're getting ready. Tourists have been warned to get out of town.

?It?s still very uncertain where the center of Isaac will come ashore in its final landfall in the northern Gulf,? Knabb told the Weather Channel on Sunday, ?and that can make all the difference as to who gets the strongest winds, who gets the strongest storm surge.

?Gradually, we?re seeing (Isaac?s) inner core develop. We?re certain it?s going to be a hurricane in the Gulf of Mexico,? Knabb said.

Cancellations and shutdowns
The Republican Party said it would recess its national convention in Tampa for a day out of safety concerns as the storm bore down. Republicans, who will formally nominate Mitt Romney as their presidential candidate for the November election, will briefly convene their four-day meeting on Monday, then recess until Tuesday.?

Gulf of Mexico oil operators braced for the first hurricane to affect the U.S. oil patch in 2012. Officials said the storm could shut down more than half of U.S. offshore oil output. Isaac's more westerly expected track brings it closer to the heart of the U.S. offshore oil patch, which produces about 23 percent of U.S. oil output and 7 percent of its natural gas output

Airlines are preparing for Isaac?s impact on Florida by waiving fees for changes and refunds for flights into and out of south Florida. Additionally, airlines on Sunday canceled most of south Florida operations.

Key West airports were closing Sunday night and most operations in and out of Miami and Fort Lauderdale were canceled until noon Monday, according to FlightAware.com. Airlines are not expecting long-term impact in Florida from the storm and should be operating normally by late Monday once airplanes, crew and staff are back in position.?

More than 740 flights to, from and within the United States were canceled Sunday in preparation for Isaac, with the bulk of the cancellations at Miami and Fort Lauderdale.

Meanwhile, the Sunday night performance of classic rock legends Lynyrd Skynyrd at Tampa?s American Action Network Pavilion at Liberty Plaza was canceled over safety concerns.

Old hat for locals
Key West locals followed time-worn storm preparedness rituals while awaiting the storm.

On Saturday, a steady line of cars moved north along the Overseas Highway, the only road linking the Florida Keys. Residents boarded up windows, laid down sandbags and shuttered businesses ahead of the approaching storm. Even Duval Street, Key West's storied main drag, was subdued for a weekend, though not enough to stop music from playing or drinks from being poured.

"We'll just catch every place that's open," said Ted Lamarche, a 48-year-old pizzeria owner visiting Key West to celebrate his anniversary with his wife, Deanna. They walked along on Duval Street, where a smattering of people still wandered even as many storefronts were boarded up and tourists sported ponchos and yellow slickers.

"Category None!" one man shouted in a show of optimism.

The Keys were bracing for storm surges of up to four feet, strong winds and the possibility of tornadoes. The island chain's two airports closed Saturday night, and volunteers and some residents began filing into shelters.

"This is a huge inconvenience," said Dale Shelton, a 57-year-old retiree in Key West who was staying in a shelter.

How to set up two-step verification for Dropbox

Popular cloud storage solution, Dropbox has begun enabling two-step verification for their accounts, which means in addition to your password (something you know), they send an additional access code to your phone, or let you generate a time-based code with a mobile app like Google Authenticator (something you own). Although no security scheme is unbreakable, two-factor authentication is absolutely better than one. And if you keep personal data in Dropbox, you're absolutely going to want to use it.

How to enable Dropbox two-step verification on your Mac or Windows PC

Dropbox is currently providing two-factor verification as a beta but will be rolling it out to all accounts over the next few days. If you're reading this during the beta period, you'll have to manually enable it by downloading the Dropbox beta from their forum.

  1. Download Dropbox 1.5.12.
  2. Install Dropbox 1.5.12.

How to enable Dropbox two-step verification on your account

Once you're sure you're running the latest, greatest, proper version of Dropbox software, you need to turn on two-factor authentication on your account.

  1. Go to http://www.dropbox.com/try_twofactor on your desktop web browser.
  2. Login to your Dropbox account if you aren't already.
  3. If everything is good to go, you'll see a green banner at the top of the web page that reads: "Success! You can now try enabling two-step verification. Look under the "Account sign in" section of this page."
  4. Scroll down to Account sign in.
  5. Two-step verification will be set to disabled, click on (change).
  6. Click on Get started at the bottom right of the Enable two-step verification pop-up.
  7. Enter your Dropbox password.
  8. Choose whether you want your verification code sent to your phone over SMS, or if you want to generate a time-based code using a mobile app like Google Authenticator.
  9. For this example, I'm using SMS, and entering my country code, area code, and iPhone number.
  10. Enter the code you received via SMS, or generated via the mobile authenticator app. (Note: The first two codes they sent me were rejected as invalid, the third one worked, so you may need to try several times.)
  11. Record your emergency backup code in case you ever lose your phone and need to temporarily disable two-step verification. (They suggest writing it on paper but I hate that so I put it in 1Password.)
  12. Click **Enable two-step verification" on the bottom right.

That's it, you're done! From now on, whenever you log in to dropbox.com or enable a new computer or mobile device, you'll have to enter the SMS or generator code to make sure everything in nice and secure.

How to login to Dropbox two-step verification on your iPhone, iPod touch, or iPad

The next time you need to add a device to your Dropbox account, for example, in a couple of months when a shiny new iPhone 5 or iPad mini follows you home from the Apple Store, you'll need to use that fancy new authentication code to do it.

  1. Download the Dropbox for iOS app if you haven't already.
  2. Launch the Dropbox app for the Home screen.
  3. Tap I'm already a Dropbox user.
  4. Enter your Dropbox account Email address and Password.
  5. Tap Log into Dropbox.
  6. Dropbox will SMS you a security code or you'll go create one using Google Authenticator. (If you're using SMS, and you're quick enough, you can watch for the Notification Center banner and type the code without ever leaving the Dropbox app.)
  7. Enter the Code.
  8. Tap Submit Code. (It only took two tries to get a valid code this time...)

That's it! Well, Dropbox will ask you if you want to enable automatic uploads (I don't, I use Photo Stream, but that's pretty much it.

What?s New: Ram pickups left the Dodge garage in 2010 to become a separate brand, and the 2013 model represents the first major makeover of the bread-and-butter half-tonner. Beneath the skin, engineers redesigned and lightened the frame and gave the 1500 a new suspension. The redesigned skin doesn?t look vastly different, but the design team claims to have created unmathed aerodynamics with a drag coefficient of 0.36. A new, more powerful 3.6-liter V-6 replaces the previous 3.7-liter as the standard engine, and a new eight-speed automatic transmission replaces the previous four-speed. The combination adds up to improvements in both performance and fuel economy.

Tech Tidbit: A subtle engineering element in the new eight-speed automatic is a thermal management system designed to raise transmission oil temperature quickly. When the engine reaches operating temps, coolant circulates through a heat exchanger to warm the trans oil, reducing its viscosity rapidly and improving fuel economy.

Driving Character: Like its competitors, the Ram is a big vehicle, and it feels big. Responses range from deliberate to slightly ponderous, and turning circles range up to 45 feet?not very handy in parking lots. The new V-6 and automatic transmission are smooth and distinctly more potent than their predecessors?Chrysler claims 0-to-60 mph in 7.5 seconds?and the Ram willingly tows up to 6500 pounds. On-road or off, ride quality is remarkably supple for such a big vehicle thanks to the new air suspension. Remarkable also applies to interior noise levels?the cabin is quiet at all speeds, on all surfaces.

Favorite Detail: The optional storage bins built into the sides of the current truck?s cargo bed (Ram Boxes) are manually lockable. But securing those three locks can be a nuisance, especially when the weather turns nasty. This inconvenience makes them a common target for thieves. The new Ram offers a power function for those three locks, integrating their operation with the power door locks. One click on the key fob secures everything.

Driver?s Grievance: The new eight-speed automatic is controlled by a dashboard rotary knob. It?s... adequate, but we?re not sure how this feature will resonate with hardcore truckers. In step with the industry, the Ram is equipped with a new electric rack-and-pinion power steering system, a trend driven by fuel economy. Although the system delivers some feedback and on-center feel in top-of-the-line models, it felt vague in the more basic version.

The Bottom Line: Besides all-new interiors and evolutionary styling?still boldly unmistakable?the new Ram stands out for four key distinctions: its new standard V-6; its new eight-speed automatic transmission; its stop/start technology; and its air-spring suspension. The combination of much improved V-6 power and efficiency, plus an all-but-seamless stop/start function, gives the Ram best-in-class EPA fuel economy: 18 mpg city/25 highway. The new V-6 overlaps the optional 4.7-liter V-8 in terms of power, and for serious punch, the 5.7-liter Hemi V-8 continues to be one of the top dogs in the half-ton pickup pack.

The Ram has been the perennial third-place player in the full-size pickup derby, but this renewal could propel it closer to the front of the field.

Two huge, exotic snakes set loose in Chesterfield County - WTVR 6

CHESTERFIELD COUNTY, Va. (WTVR)?Two expensive, exotic South American snakes were set loose in Chesterfield County.

Last night CBS 6 told you about a red-tailed boa constrictor discovered in Chesterfield County.? Today police recovered another super-sized serpent has been recovered, this one an albino Burmese python.

Authorities now believe that all reptiles are accounted for, and Rob Leinberger, with Animal Control said that ?all evidence that we found so far led us to believe that there were probably two snakes.?

Animal control officers went door-to-door Wednesday morning, searching for the second snake after a boa constrictor was found coiled up Monday in the storage area at One Hour Heating and Air Conditioning.

The python was also recovered in the woods line off East Boundary road, actively roaming around. It was taken to a local exotic pet shop.

?It was probably man-handled the past couple of day, and she?s very nice, tongue coming out, she?s moving, she?s curious, she?s healthy?she?s like she is supposed to be,? said Vinny Ferraiola, owner of the exotic pet store. ?There is nothing evil about her.?

Officers said they did find evidence that the boa and python were released in a wooded area behind the complex. They?re now trying to figure out who owned the snakes.

The python will be housed there for the next month.

?We have to keep her for 30 days as evidence, other than that the market for this animal is huge,? said Ferraiola. ?She?s a pretty animal?probably breedable next year.?

Animal Control asks viewer to contact them if you know anyone that had two pet snakes, specifically a boa and python, and they no longer have them.

That office number is 748-1683. Officers are 99 percent sure these expensive snakes were pets and are trying to figure out why they were released into the wild.

Native American spiritual beliefs influential in spurring youth to avoid drugs and alcohol

ScienceDaily (Aug. 20, 2012) ? New research indicates that urban native American youth who follow American Indian traditional spiritual beliefs are less likely to use drugs and alcohol. Arizona State University social scientists will present their findings at the 107th Annual Meeting of the American Sociological Association in Denver, Colorado.

The study, "Spirituality and Religion: Intertwined Protective Factors for Substance Use Among Urban American Indian Youth," was recently published in The American Journal of Drug and Alcohol Abuse. The authors are: Stephen Kulis, the study's principal investigator and ASU School of Social and Family Dynamics professor; David R. Hodge, ASU School of Social Work associate professor; Stephanie L. Ayers, ASU Southwest Interdisciplinary Research Center associate director of research; Eddie F. Brown, ASU American Indian Studies professor and American Indian Policy Institute executive director; and Flavio F. Marsiglia, ASU School of Social Work professor.

"Most American Indians now live in cities rather than tribal communities. Our study is one of the few to address the role of spirituality and religion among urban Native youth, recognizing the unique histories of cultural integration that characterize today's urban American Indian communities and the complex belief systems and practices that sustain them in the urban landscape," Kulis said.

Among the general native American youth population, higher rates of substance (both drug and alcohol) abuse are reported than among their non-native American counterparts. They also are more likely to use heavier amounts, initiate substance use earlier, and have more severe consequences from substance use, according to past research.

Native Americans typically do not separate spirituality from other areas of their lives, making it a complex, cultural and intertwined aspect of their daily existence.

Researchers found that adherence to native American beliefs was the strongest predictor of anti-drug attitudes, norms, and expectations. Concerning substance use, aspects of spirituality and religion associated with lower levels of use were affiliation with the Native American Church and following Christian beliefs.

Data for the study were collected from native American students enrolled in five urban middle schools within a large southwestern city in 2009. The average age of the 123 respondents was 12.6 years old.

Most of the study respondents expressed strong anti-drug and alcohol beliefs, with the majority stating that they "definitely would not" use alcohol, cigarettes, or marijuana if given an opportunity (55 percent) and that it was "definitely not okay" for students their age to use those substances.

Respondents also felt that their parents (78 percent) and grandparents (69 percent) would be "very angry" if they used drugs or alcohol and 51 percent stated they were "very sure" that they would reject any substance offers.

Also notable was that about half (53 percent) had resisted offers of drugs in the past 30 days.

Spirituality was reflected as an important aspect in students' lives. More than 80 percent of respondents said that spirituality held some importance to them and was part of their lives. Seventy-nine percent of the students felt it was "somewhat" or "very important" to follow traditional American Indian beliefs and about half felt it was important to follow Christian beliefs.

However, a general sense of spirituality that did not refer specifically to native American traditions, beliefs, or culture was not found to be a deterrent against substance use.

"Rituals and ceremonies have helped American Indian communities adapt to change, integrate elements of different tribes, infuse aspects of Western organized religions, and make them their own," according to the paper.

In addition, the paper states that possessing a feeling of belonging to traditions from both American Indian and Christian cultures may foster integration of the two worlds in which urban American Indian youth live.

Syrian FM claims rebels no match for military

BEIRUT (AP) ? Syria's foreign minister defiantly dismissed rebel forces and their international backers on Thursday as incapable of toppling the military defending Bashar Assad's regime, even as condemnation grew over expanded offensives that activists say have claimed dozens of civilian lives in recent days.

Foreign Minister Walid al-Moallem's confident tone contrasted sharply with a series of recent blows to Assad, including high-level military and political defections and the ability of rebel guerrillas to stage bombings and abductions in the heart of the capital, Damascus.

The timing of al-Moallem's interview on Syrian state TV also suggested attempts to reassure Assad's supporters at a time when Damascus has few reliable allies remaining. Iran stands firmly behind Assad, but the critical bonds are with U.N. Security Council members China and Russia, which have blocked efforts to impose sanctions and other measures to pressure Syria.

A Syrian envoy, Bouthaina Shaaban, was in Beijing on Thursday and described talks with China's foreign minister as "really great."

"Those who think that the Syrian Arab army will be defeated are dreaming," al-Moallem said.

He also repeated Syria's strong denunciations of key rebel backers, such as Qatar and Saudi Arabia ? main rivals of Iran that would like to undercut Tehran's most important alliance in the region. Syria has become perhaps the most complex proxy stakes of the Arab Spring, with Gulf Arab states, NATO member Turkey and the West working to weaken Assad's regime.

The U.N. is groping for ways to remain relevant as violence has all but eclipsed efforts for a peace plan. A U.N. military observer mission ends Sunday and will be replaced by a new civilian office to try to push ahead diplomatic bids to end the more than 17-month civil war, which activists say has left more than 20,000 people dead and forced hundreds of thousands from their homes.

A U.N. report on Wednesday said war crimes have been committed on both sides, including by Syrian government forces and allied militiamen blamed for the deaths of more than 100 people, nearly half of them children, in the village of Houla in May.

During a visit to a refugee camp in Jordan, French Foreign Minister Laurent Fabius assailed Assad for "butchering his own people."

"The sooner he goes the better," he said.

Syrian forces, however, have stepped up campaigns to drive back rebel advances across Syria. One of the main battlegrounds is the country's largest city, Aleppo, where some areas have been pounded by weeks of government shelling and airstrikes.

Another struck Thursday just after dawn, targeting a bread line outside a bakery in Aleppo and killing at least 10 people, activists said.

Mohammad al-Hassan, an Aleppo-based activist who saw the aftermath of the attack, told The Associated Press by telephone that the shells hit at 6:30 a.m. when most people line up for bread ? a staple that is running in short supply ? before the day gets too hot.

"Three shells hit the street near the bakery and people who had been waiting were hit by shrapnel," he said. "There were people with their children there. It was like a river of blood."

Al-Hassan said he saw dead bodies on the pavement and also spoke to witnesses at the scene. The two main activist groups, the Britain-based Syrian Observatory for Human Rights and the Local Coordination Committees, confirmed the details of the attack. The Observatory said 20 people were killed in shelling of Aleppo on Thursday, including at least 10 outside the bakery.

A video posted by activists online showed the bloodied bodies of at least two young children covered with blankets and the corpses of three men soaked in blood that ran onto the pavement. The authenticity of the video could not be independently verified.

On Wednesday, warplanes exacted a heavy toll with airstrikes on a residential neighborhood in the rebel-held town of Azaz close to the Turkish border. International watchdog Human Rights Watch said more than 40 people were killed and at least 100 wounded, many of them women and children. AP reporters saw nine bodies in the bombings' immediate aftermath, including a baby.

The strikes leveled entire blocks in a poor neighborhood and sent panicked civilians fleeing for cover. So many were wounded that the local hospital locked its doors, directing residents to drive their injured to the nearby Turkish border for treatment on the other side.

"This horrific attack killed and wounded scores of civilians and destroyed a whole residential block," said Anna Neistat, acting emergencies director of Human Rights Watch. "Yet again, Syrian government forces attacked with callous disregard for civilian life."

Azaz, which is home to around 35,000 people, is also the town where rebels have been holding 11 Lebanese Shiites they captured in May. A series of hostage-takings by rebels ? including grabbing a member of a powerful Lebanese Shiite clan ? has touched off retaliatory abductions of Syrians in neighboring Lebanon and raised worries about the country being dragged into deeper unrest.

Lebanon is a volatile mix of pro- and anti-Syrian factions ? many of them armed, such as the formidable Assad ally Hezbollah.

In Lebanon's eastern Bekaa Valley, near the Syrian border, masked men believed to be backing the Syrian rebels set up roadblocks on a main highway. In a separate incident, gunmen attacked a car driven by a pro-Syrian Lebanese politician, Joseph Abu-Fadel, breaking his car windows with stones as he was driving to Syria. He and three others were slightly injured.

Lebanese military units set up along the main airport highway in Beirut to avoid another protest by pro-Assad mobs, which blocked the road Wednesday and forced passengers to walk to catch their flights.

In Damascus, U.N. humanitarian chief Valerie Amos said the Syrian conflict has "become more intense and is too often indiscriminate" and estimated as many as 2.5 million people are in need of relief assistance.

Later, in Beirut, Amos expressed frustration at Syria's reluctance to allow more major international aid groups into the country because of Syrian fears that relief supplies could reach rebels."They don't want to see that happen," she said.

In another symbolic blow to Syria, the Organization of Islamic Cooperation suspended Syria from the group during a meeting in Saudi Arabia. The move brought a swift denunciation from Iran, Assad's main regional ally.

Iran's foreign minister, Ali Akbar Salehi, criticized the decision as "unfair" because Syria was not invited to the Mecca summit, which wrapped up early Thursday.

Blocking destruction of defective proteins unexpectedly delays neurodegeneration in mice

ScienceDaily (Aug. 15, 2012) ? One might expect that ridding a brain cell of damaged proteins would be a universally good thing, and that impairing the cell's ability to do this would allow the faulty proteins to accumulate within the cell, possibly to toxic levels. So a lot of scientific effort has gone into looking for ways to enhance the process by which cells dispose of banged-up proteins.

But this thinking may need some revision, according to a new study from the Stanford University School of Medicine. Senior author Thomas Sudhof, MD, professor of molecular and cellular physiology, and his fellow researchers have unexpectedly found that inhibiting the process by which damaged proteins are ordinarily broken down within cells both delayed the onset of symptoms in laboratory mice that are highly prone to neurodegeneration and significantly increased their longevity.

The study, published online Aug. 15 in Science Translational Medicine, also showed that blocking the activity of cells' in-house garbage disposals -- known in the biology business as proteasomes -- in nerve cells taken from the neurodegeneration-prone mice restored several key biochemical characteristics necessary for the cells' healthy function.

Although Sudhof cautions that more research is needed, the findings pose a challenge to prevailing beliefs about the pathology of certain neurodegenerative disorders.

"The current consensus in neuroscience favors a therapeutic strategy of trying to accelerate, rather than impede, the disposal of damaged proteins that accumulate in the brains of patients with Alzheimer's, Parkinson's, Huntington's and other neurodegenerative diseases," said Sudhof, who is also the Avram Goldstein Professor in the School of Medicine.

Proteasomes are cell components that destroy damaged proteins. Not just nerve cells but virtually all cells in creatures ranging from yeast to humans contain multitudes of these tiny tube-shaped machines, which suck the defective proteins into their holes and chop them into smithereens.

Examining brain tissue from deceased Alzheimer's and Parkinson's patients, Sudhof's team observed that an aspect of the degenerative process identical to the one they had prevented in lab mice was occurring, suggesting that their findings in the animal model might prove relevant to these and other human diseases as well.

The particular defective protein whose demolition was staved off in Sudhof's study, called SNAP-25, plays a key role in the release of chemical signals that nerve cells use to communicate with one another. But like all proteins, SNAP-25 can't do its job until it is slapped into shape.

"Structure equals function" is a watchword of biochemistry. Proteins -- the molecular creatures that do the bulk of the work in every living cell -- are initially produced as long linear sequences of small chemical subunits that progressively get strung together like beads on a string. But the string is just a string until it assumes a specific structure, typically with help from one or more "chaperone" molecules that midwife it into its correct conformation.

Like a mail carrier's feet, overworked proteins can eventually go flat. Misfolded and therefore no longer functional proteins may, alternatively, be quickly reconditioned and put back on the job or get chemically "barcoded" for demolition at the hands of proteasomes.

SNAP-25 is used by many types of cells but works particularly hard in nerve cells, or neurons. To transmit signals to one another, neurons release specialized chemicals into small gaps called synapses that separate one neuron from the next in a relay. Prior to release, those chemicals are sequestered within membrane-bound packets, or vesicles, inside the neuron. Every time one neuron transmits a signal to the next -- which can be more than 100 times a second -- hundreds of tiny chemical-packed vesicles approach the edge of the first neuron and fuse with its outer membrane. A fused vesicle's inner surface becomes part of the neuron's outer surface (just as would happen if a small bubble merged with a larger one surrounding it), and its stored contents spill out into the synapse.

To make this happen, vesicles must be actively coerced into close contact with neurons' membranes. This is accomplished by a cluster of proteins that serves as the molecular equivalent of a clamp. In the course of repeated bouts of neuronal firing, SNAP-25 -- a key constituent of this clamping complex -- can get bent out of shape, rendering the entire clamp assembly useless. The more a neuron fires, the more molecules of SNAP-25 get deformed over time.

Some years ago, Sudhof's group used a sophisticated genetics technique to create a strain of laboratory mice lacking a chaperone molecule that assists in the proper refolding of SNAP-25. These mice seem quite normal early in life. But the gradual failure of the clamping action necessary for vesicle fusion and neuronal signaling causes the neurons to die off, eventually triggering behavioral symptoms and early death. Sudhof has used these mice extensively as a model system for understanding misfolded proteins and degenerative disease.

In the new study, giving the chaperone-deficient mice injections of either of two proteasome-inhibiting drugs (lactacystin and epoxomicin) once every five days -- probably far from an optimal regimen, but at least a feasible animal experiment, Sudhof said -- delayed typical outward symptoms of their neurodegenerative disorder by as much as 30 percent and prolonged their survival by close to 20 percent. Next, the researchers incubated neurons from the experimental mice in solutions with or without proteasome-inhibiting drugs. Biochemical measurements showed that, while overall stores of SNAP-25 in the neurons incubated with or without drugs were the same, the activity of the clamping complex was restored to normal levels in the drug-soaked neurons. (In the drug-free neurons, clamp activity remained hugely impaired.)

Clearly, preventing the breakdown of misfolded SNAP-25 molecules increased their levels in neurons -- an outcome that one would intuitively think would be toxic, Sudhof said. But the experiments in the new study instead suggest that misfolded proteins, either randomly just from being bounced around in the surrounding environment or with the active assistance of "generic" chaperone molecules, can revert to normal shapes and jump back into the fray. As faulty SNAP-25 accumulates in the cell, more of it gets repaired, with beneficial downstream implications for molecular-clamp activity, vesicle-membrane fusion and better chemical signaling by neurons. Because neurons have to signal effectively in order to survive, fewer of them die.

A number of neurodegenerative disorders such as Alzheimer's, Parkinson's and Huntington's diseases are associated with the occurrence of damaged proteins. It is a widely held view among investigators in the field that the buildup of these damaged proteins substantially contributes to the disease process.

Sudhof cautioned that, while it is possible that proteasome-inhibiting drugs could, paradoxically, turn out to slow the progress of these and other human degenerative conditions, this was far from proven. However, far more powerful proteasome-inhibiting drugs than the old, off-patent drugs Sudhof and his teammates gave their experimental mice are already in clinical use for certain cancer indications, so they could rapidly be deployed in preclinical studies testing this possibility.

World Muslim leaders mull suspending Syria

Muslim leaders were on Tuesday to mull proposals to suspend war-hit Syria from the 57-member Organisation of Islamic Cooperation (OIC), an action which is strongly opposed by Iran.

The OIC represents 1.5 billion Muslims worldwide and that it is holding a meeting Tuesday night in Mecca, the holiest Islamic city, is of particular significance -- the 26th of the holy month of Ramadan is the night when the Koran was revealed to the Prophet Mohammed, according to Muslim tradition.

Foreign ministers who held a preparatory meeting on Monday recommended the suspension of Syria's membership, the OIC chief Ekmeleddin Ihsanoglu said.

Iranian President Mahmoud Ahmadinejad, whose country has openly criticised the push to suspend Syria, is attending the extraordinary meeting.

It was proposed by Saudi King Abdullah, who is pushing to mobilise support for the Syrian rebels.

Tensions had been simmering for months between Sunni-dominated Saudi Arabia and Shiite-dominated Iran, as both long-standing regional rivals had taken opposite stances on the uprising in Syria, which topped the summit's agenda.

The United States said that Rashad Hussain, its special envoy to the OIC, would take also part in the Mecca summit as an observer and hold meetings with other delegates on the sidelines.

His attendance "demonstrates the United States' commitment to working with our partners in the international community to support the aspirations of the Syrian people and bring additional pressure to bear on the Assad regime," a State Department statement said Monday.

Iran is the Syrian regime's biggest regional ally and has pledged its full support for embattled President Bashar al-Assad in his fight to remain in power, though it denies providing him with soldiers or arms.

Earlier this month, ISNA news agency quoted Iranian Foreign Minister Ali Akbar Salehi as saying that "retired" members of Iran's Revolutionary Guards and army are among 48 Iranians taken hostage by rebels in Syria.

Tehran, however, accuses Saudi Arabia, Qatar and Turkey of arming and financing the mainly Sunni Syrian rebels against Assad, who hails from the Alawite minority -- an off-shoot of Shiite Islam.

Meanwhile Saudi Arabia, which hosts the headquarters of the OIC based in the Red Sea city of Jeddah, has openly called for arming Syrian rebels.

Salehi on Monday assured reporters that "we are on track for increasing and improving the level of relations with Saudi Arabia."

However, the Iranian minister criticised the move to suspend Syria's membership of the OIC.

"We certainly do not agree with the suspension of any OIC member," Salehi said after the meeting.

"The suspension of its membership does not really resolve the issue and is not in line with the OIC charter," he said. "We have to look for other ways, means and mechanisms for resolving conflicts and crises."

Salehi called for "paving the way for a meeting between the opposition and the Syrian government so they can negotiate with the help of others to reach a Syrian-Syrian solution."

But apparently, most OIC members disagreed with Iran.

"The decision has been agreed upon based on consensus with an absolute majority" in favour of suspending Syria's membership, Ihsanoglu said.

It will be put to heads of state at their summit in Mecca, which will continue on Wednesday, for "final approval," he added.

Sources close to the meeting said that only Iran and Algeria were against the recommendation. Syria had no representatives at the meeting.

Meanwhile, Mohammed Ahmed Taieb, a top Saudi foreign ministry official told AFP that "some delegates" were calling for further steps against Damascus by "demanding that the current president step down and preparing for a post-Assad transition period."

Tunisian Foreign Minister Rafik Abdessalem hailed the suspension as "a strong message to the Syrian regime on the importance of listening to the will of the people and their demands for freedom, justice and dignity."

The 17-month conflict in Syria has killed more than 21,000 people, according to monitoring groups.

In addition to the Syrian crisis, the leaders were also to discuss the Arab-Israeli conflict, the violence against the Muslim Rohingya minority in Myanmar, as well as the unrest in Mali.

On the lighter side, here?s a critique on our ?stand-up? routine at the Market Research in the Mobile World Conference.

AP ? After being generously introduced by Master of Ceremonies, Lenny Murphy, as the funniest man in market research, KL Communications? CEO Kevin Lonnie spent the next ten minutes being spectacularly unfunny!

As if to completely rebuff his gracious introduction, Mr. Lonnie?s highly anticipated comedy routine quickly dissolved into a ?dry as toast? PowerPoint presentation.

This author was not alone in his opinions of this atrocious comedy act.

Mr. Gregg Archibald, Senior Partner of new consulting firm Gen 2 described it this way.? ?After Lenny?s intro, I was ready to burst into laughter at the 1st sign of something remotely humorous.? But alas, that moment never came.?

Echoing Mr. Archibald?s lament, Mr. Ricardo Lopez, CEO of Hispanic Research Inc. described it this way, ?As a presentation it was OK, but it wasn?t the least bit funny.?

Fortunately, no chants for ?Refund? emerged from the audience as Mr. Lonnie had the good sense to quickly surrender the stage to the far more loquacious Sean Holbert.

The audience was further aided by the 20 minute presentation limit upon which Mr. Murphy quickly gave the hook to ?the funniest man in market research? (an oxymoron if ever there was one. ).

As for this author, he has serious doubts that Mr. Lonnie?s comedic career can resurface.? ?This was a burn out that comes along once a decade, it?s our generation?s answer to Howard The Duck? said an unnamed conference registrant.

Mr. Lonnie?s booking agent [name withhold for obvious reasons] said that Mr. Lonnie would be back on stage at QRCA?s Annual Conference in Montreal this October.? Attendees of that event are advised to find their own source of amusement during Mr. Lonnie?s next ?performance?.

'Strawberry' birthmarks grow rapidly when babies just weeks old

ScienceDaily (Aug. 14, 2012) ? Strawberry-shaped birthmarks called infantile hemangiomas grow rapidly in infants much earlier than previously thought, Mayo Clinic and University of California, San Francisco, researchers found. Their study, published online in the journal Pediatrics, suggests that babies with complication-causing hemangiomas should be immediately referred to dermatologists for further evaluation.

Infantile hemangiomas are the most common tumor in infancy. They tend to appear in the first weeks of life and grow as a child ages. Potential complications include permanent disfigurement of the face or functional compromise of vital organs.

"Our goal was to try to figure out when this actual period of rapid growth happened," says Megha Tollefson, M.D., a Mayo Clinic Children's Center researcher and pediatric dermatologist who conducted the study with Ilona Frieden, M.D., of the University of California, San Francisco.

"Then we could potentially intervene if we had to."

The researchers examined photos of 30 infants from birth to 3 months, analyzing the color, thickness and distortion of anatomic landmarks.

Previously, physicians believed that the tumors grew during the first 5 months of life, but researchers had not yet discovered when the most rapid growth took place.

"By using a novel study design, we were able to demonstrate that the period of most rapid hemangioma growth of superficial hemangiomas occurs between 5.5 and 7.5 weeks of age," Dr. Frieden says.

The new findings suggest that infants with high-risk infantile hemangiomas should be seen by a dermatologist as soon as possible, preferably by 4 weeks old. This way therapy, such as drug treatment and laser removal, can start as soon as possible.

"Depending on where the hemangioma is located, it could potentially have long-term impact," Dr. Tollefson says. "We now have the possibility of preventing a lot of that."
